Antibiotic prophylaxis in orthopedic surgeries: the results of an implemented protocol BJID
Queiroz,Raquel; Grinbaum,Renato S.; Galvão,Luciana L.; Tavares,Fernando G.; Bergsten-Mendes,Gun.
Though the basic principles of antibiotic prophylaxis have been well established, there is still considerable incorrect usage, including how much is prescribed and especially in the duration of treatment, which is generally superior to what is indicated. The adequate use of these drugs contributes towards decreasing the time of internment of the patient, prevents surgical site infection (SSI), decreasing the development of resistant microorganisms, and towards reduced costs for the hospital pharmacy. A protocol for the use of antibiotic prophylaxis in the Orthopedics and Traumatology Service of the Hospital do Servidor Público Estadual de São Paulo was developed. The objectives of the study were to promote rational antibiotic surgical prophylaxis, through...

Conocimiento tradicional y cultivo in vitro del cuatomate (Solanum glaucescens Zucc). Colegio de Postgraduados
Medina Galicia, Alfonso.

CTAB methods for DNA extraction of sweetpotato for microsatellite analysis Scientia Agricola
Borges,Aline; Rosa,Mariana Silva; Recchia,Gustavo Henrique; Queiroz-Silva,Jurema Rosa de; Bressan,Eduardo de Andrade; Veasey,Elizabeth Ann.
Microsatellite markers have proved to be useful in genetic diversity assessments of sweetpotato (Ipomoea batatas) but practical DNA extraction methods to ensure good quality and quantity DNA for these studies are yet to be established. This study compares the efficiency of three modified methodologies for DNA extraction of six sweetpotato landraces using the CTAB extraction buffer in regard to quantity and purity of DNA quantification and microsatellite band patterns. All methodologies yielded satisfactory results, but the method based in leaf tissue macerated in liquid nitrogen was deemed more adequate because of its simplicity and lower cost. However, the method based in dry leaf tissue was considered more advantageous, first because elicits...

Effects of extraction method and storage of dry tissue on marine lipids and fatty acids ArchiMer
Sardenne, Fany; Bodin, Nathalie; Metral, Luisa; Crottier, Anais; Le Grand, Fabienne; Bideau, Antoine; Brisset, Blandine; Bourjea, Jerome; Saraux, Claire; Bonhommeau, Sylvain; Kerzerho, Vincent; Bernard, Serge; Rouyer, Tristan.
Various protocols are currently used to study marine lipids, but there is a growing interest in working on dry samples that are easier to transport. However, reference protocols are still lacking for dry samples. In order to make recommendations on this use, lipid classes and fatty acids (FA) obtained from six analytical protocols using two different tissue states (dry vs wet) and three extraction methods (automat vs manual potter vs leaving the solvent to work on tissue) were compared. Three dry storage modes of tissue (freezer vs gas nitrogen vs dry room) during one and three months were also compared. These comparisons were made on seven marine species with different lipid profiles, including fishes, crustaceans and mollusks. Lipid classes and FA...

Evaluation of Free-Range broilers using the welfare quality® protocol Rev. Bras. Ciênc. Avic.
Sans,ECO; Federici,JF; Dahlke,F; Molento,CFM.
Scientific information on the welfare of broilers reared in alternative systems is scarce. The objective of this study was to assess the welfare of free-range broilers using the Welfare Quality® protocol. Free-range broilers reared in ten farms were observed, and measures were made in broilers of five of these farms. The collected data were transformed into scores (0-100), with higher scores indicating better welfare, except for emotional states. Prevalence percentages were calculated for the remaining data. Median (min-max) scores were 81 (63-98) for lameness, 93 (83-99) for hock burn on the farm, 100 (95-100) for plumage cleanliness, 35 (8-70) for podermatitis on the farm, 56 (26-88) for density, 53 (20-53) for dust, 34 (14-67) for litter quality, 93...

Identification et quantification des pigments chlorophylliens et caroténoïdes des sédiments marins : un protocole d'analyse par HPLC ArchiMer
Brotas, V; Plantecuny, Mr.
A HPLC protocol is described which permits analysis of the pigment composition of coastal and intertidal sediments. The HPLC technique has been extensively applied to phytoplankton pigments for fifteen years. Papers dealing with living benthic microphytic populations have been more rare, due to complexity existing in the sediment surface, where pigments (chlorophylls and carotenoids) from several taxonomic groups of microalgae are mixed with degraded pigments from senescent phytoplankton cells and macrophytic material. The present paper describes in detail a protocol adapted from Kraay et al. (1992), which allows a good discrimination of photosynthetic pigments of microphytobenthos communities. The following procedure is recommended. Sediment samples are...

Indicators to assess goat welfare on-farm in the semiarid region of Brazilian Northeast Ciência Rural
Leite,Luana Oliveira; Stamm,Fabiana de Orte; Garcia,Rita de Cassia Maria.
ABSTRACT: In 2015, AWIN Goat Protocol was published in Europe, with indicators that identify welfare level of lactating dairy goats; however, there are no welfare protocols for meat goat. Therefore, the objective of this study was to select animal and resource-based indicators to assess welfare in meat goat in different types of grazing systems in Brazilian semiarid Northeast. Eighteen indicators were selected to evaluate the welfare of meat goat. The knowledge of the welfare degree of these animals is the best way to make improvements and promote a better quality of life to meat goat.

Isolamento de DNA genômico de indivíduo de nematoide parasita de insetos: estudo de caso de Deladenus PFB - Pesquisa Florestal Brasileira
Schuhli, Guilherme Schnell e; Penteado, Susete do Rocio Chiarello; Kestring, Daiane Rigoni; Steil, Gisleine Jarenko; Thomas, Micheli Cristina.
Populational studies involve single individual DNA extraction in order to grant further genotyping data. In the case of some parasitic nematodes, the reduced dimensions and high individual number per infestation makes individual genotyping a difficult task. Aiming the development of a protocol we performed adjustments in available methods in order to acquire the best gain in purity and concentration of genomic DNA. Single specimens were digested in Worm Lysis Buffer and submitted to PCR amplification as a concept test. It was possible to obtain good amount and concentration of DNA from individuals. Quality was sufficient to grant subsequent ITS1 sequencing.

Model of induction of thyroid dysfunctions in adult female mice Arq. Bras. Med. Vet. Zootec.
Ferreira,E.; Silva,A.E.; Serakides,R.; Gomes,A.E.S.; Cassali,G.D..
It is described the elaboration of a protocol to induce hyperthyroidism and hypothyroidism in mice by administrating thyroxin and propylthiouracil, respectively, in the drinking water. The drugs were administered to adult female mice of the Swiss strain for 30 days in order to obtain a systemic status of thyroid dysfunction. The induction of hyperthyroidism and hypothyroidism in the animals was confirmed by the histomorphological analysis of the thyroid in the end of the experiment, when the state of gland dysfunction in the animals submitted to the treatment was observed.

Molecular analysis of genetic stability in long-term micropropagated shoots of banana using RAPD and ISSR markers Electron. J. Biotechnol.
Lakshmanan,Venkatachalam; Reddampalli Venkataramareddy,Sreedhar; Neelwarne,Bhagyalakshmi.
A large number of micropropagated plantlets of banana, Musa acuminata var. Nanjanagudu Rasabale (NR), that were developed from axillary shoot bud explants over 10 years ago were screened for genetic variation, if any, using RAPD (Random Amplified Polymorphic DNA) and ISSR (Inter-Simple Sequence Repeats) markers. Of the 4000 in vitro plantlets, 11 were used for screening that involved shoot cultures with distinct variation in morphological characteristics (morphotypes). Similarly, the mother maintained in the field was also subjected for genetic analysis. Out of the 50 RAPD and 25 ISSR primers screened, 30 RAPD and 5 ISSR primers produced totally 424 clear, distinct and reproducible band classes resulting in a total of 5088 bands where the banding patterns...
